The song is from the album ‘Ritu Hirwa Marathi Bhaavgeete’ and the beautiful lyrics are penned by B. Many of the laboratory procedures involve the use of dilutions. Serial dilution is a simple yet efficient technique to determine the number of cells or organisms in a concentrated. As example, if 1.0 ml of 3M HCl is added with 9 ml of distilled water, the dilution factor becomes 1/10. Serial Dilution: Importance and Aplication. Then, 1 mL from that mixture is added to 9 mL, and it is mixed together (a 10-2 dilution). As shown in the illustration below, it begins when 1 mL of the bacterial sample is added to 9 mL, and it is mixed together (creating a 10-1 dilution). The factor can be obtained by dividing the actual volume of the initial chemical used to the final volume of the chemical after water is added. A serial dilution is the dilution of a sample, in 10-fold dilutions. Ideally, therefore, we would like to get 100 bacteria on the plate that we count. A ten-fold serial dilution could be 1 M, 0.1 M, 0.01 M, 0.001 M. We would like to count the bacteria in a particular sample, and we think that there should be in the neighborhood of 10 million cells per mL. Usually the dilution factor at each step is constant, resulting in a geometric progression of the concentration in a logarithmic fashion. One way is by obtaining a factor called the Dilution Factor (DF). A serial dilution is the stepwise dilution of a substance in solution. After performing the dilution, we need to know how much difference are the diluted chemical and the initial, undiluted ones. Cultures are usually allowed to grow through a normal growth curve, with daily transfer of a small volume of the expanded culture into fresh medium. |
